three, b = 65.8, c = 97.2??. Inspection of the correlation coefficient for half-data-set intensities, as recommended by Karplus & Diederichs (2012 ) and Evans (2012 ), suggests that the diffraction selleck compound information extend to a resolution of d min = 1.46?? with an overall R merge of 6.1% and an R meas of 6.7% (for details, see Table 1 ). By using the method of Matthews (1968 ), as implemented by Kantardjieff & Rupp (2003 ), it was estimated that the crystals contained a single PBGD monomer per crystallographic asymmetric unit with a solvent content of 53%. Structure analysis by use on the molecular-replacement program MOLREP (Vagin & Teplyakov, 2010 ) with E. coli PBGD (47% identity; PDB entry 1pda; Louie et al., 1992 ) as the search model was successful and refinement of the B.

megaterium PBGD structure is currently in progress using this high-resolution data set. Figure 4 A diffraction image of B. megaterium PBGD obtained on DLS beamline I03 using thing a?PILATUS 6M-F detector. The diffraction information obtained from this crystal were processed to a resolution of 1.46??. Table 1 Data-collection and processing statistics for B. megaterium PBGD Acknowledgments We gratefully acknowledge the Pakistan Higher Education Commission for a scholarship to NA. We acknowledge the Diamond Light Source (DLS, UK) for beam time and travel support (award No. MX-7131). We thank Dr Graham Taylor [Centre for Amyloidosis and Acute Phase Proteins, UCL Division of Medicine (Royal Free Campus)] for assistance with mass spectrometry.
Carbonic anhydrases (CAs) are mainly zinc metalloenzymes that catalyze the reversible hydration of carbon dioxide to bicarbonate and a proton (Krishnamurthy et al.

, 2008 ). CAs are ubiquitously expressed and are found in almost all living FERMT2 organisms, where they play a vital role in various physiological processes. Such processes include respiration, pH regulation, CO2 fixation, photosynthesis and ion transport (Supuran, 2008 ). There are five evolutionary and structurally distinct classes of CA: -CA (expressed in vertebrates and the only class found in mammals), -CA (found in plants, fungi and prokaryotes), -CA (found only in archaebacteria and diatoms) and -CA and -CA (found in diatoms) (Aggarwal et al., 2013 ). Depending over the coordination on the Zn2+ during the active site, --CAs are divided into two subclasses or clades designated type I and type II (Lotlikar et al.

, 2013 ). Type I -CAs have an open active site with a Zn2+ ion tetrahedrally coordinated by two Cys residues, one His residue and a water molecule for catalysis, and show activity both above and below pH eight (Smith & Ferry, 1999 ; Covarrubias et al., 2005 , 2006 ; Nishimori et al., 2010 ). Type II -CAs have a closed active site together with the Zn2+ coordinated by a His, an Asp and two Cys residues (Cronk et al., 2001 , 2006 ; Covarrubias et al.
